SIGNIFICIANCE OF BROMODEXYURIDINE(BRDU) LABELING INDEX IN LARYNGEAL CRACINOMA





Abstract
Bromodeoxyuridine(BrdU), an analogue of thymidine, can be detected by means of monoclonal antibodies and. utilized as a marker of the S-phase of the cell cycle. We assessed BrdU-Iabeling index(LI) from laryngoscopic biopsy specimens of laryngeal cancer as a part of their initial evaluation scheme to measure the proliferative activity of the tumor cells and to compare these results with other variables such as histopathological differentiation, site and size of the tumor. Twenty fresh biopsy specimens (well differentiated : 6 cases, moderately differentiated: 12 cases, poorly differentiated: 2 cases) were obtained. A tiny piece of tissue was acquired from the biopsy specimen and further divided into 1mm pieces and incubated in RPM1 164() with 10 u l of BrdU(600 u g/ml) and 10 u 1 of conc. H2O2 using the BrdU incubator. Immunohistochermcal staining was performed with anti-BrdU antibody and the Iabeling index was assessed with an image analyzer. The mean BrdU-LI was 12.86.9 in moderately differentiated cancer and 2.2 2.2 for well differentiated cancer. Poorer differentiation correlated with higher BrdULI(p<0.05). However, no correlation was found between the BrdU-LI and the site or size of the tumor bulk. The authors conclude that BrdU-LI can be utilized as an analogue to histopathological differentiation for further evidence in predicting the aggressiveness of the tumor.
